Abstract: | This work investigates into the tailor made generation of functional particles by means of supercritical antisolvent (SAS) processing. The supercritical antisolvent technology is utilized to generate particles respectively particulate systems with defined properties. In its plainest mode, a liquid solution of a solvent and a solute is injected into supercritical carbon dioxide which acts as an antisolvent. This leads to a supersaturation of the solute which is compensated by nucleation and particle growth. The disperse properties of the produced particles can strongly be influenced by varying the process parameters.
The aim of this work is therefore to further investigate into a possible differentiation criterion between precipitating and crystallising solutes, based on their saturation solubility in mixtures of solvent and antisolvent. Furthermore the main influencing parameters of the respective SAS particle generation models that can be utilized to tailor the final properties of the resulting particles are examined. This is done on behalf of model compounds that can be either micronized separately or processed together in the form of so-called solid dispersions. The detailed investigation into the properties of the singular compounds will be considered to validate the discussed models and even extend it to further variables. The final step of this thesis is the combined processing of the two different kinds of solutes in the form of solid dispersions with a focus on the generation of functional particulate systems with tailored properties for pharmaceutical applications. |
