THCA Synthase from Cannabis sativa L. – a biocatalyst for the production of cannabinoids
Die vorliegende Arbeit befasst sich mit dem Potential und dem zellfreien Einsatz der FAD-abhängigen Oxidase Δ9-Tetrahydrocannabinolsäure Synthase (THCAS), welches ein Enzym aus dem Sekundärmetabolismus der Cannabis sativa L. Pflanze ist. THCAS katalysiert die Bildung einer intramolekularen C-C Bindung von Cannabigerolsäure (CBGA) in Δ9-Tetrahydrocannabinolsäure (THCA), wobei Sauerstoff als finaler Elektronenakzeptor dient. Im Vergleich zu Escherichia coli erwies sich Pichia pastoris als vorteilhafter Wirtsstamm für die funktionale Genexpression des Enzyms. Die Produktion des Enzyms im prokaryotischen Wirt E. coli resultierte in der Bildung großer Mengen von Protein, jedoch in Form von unlöslichen und inaktiven Inklusionskörpern. Refolding-Versuche waren nicht erfolgreich. Aus dem Überstand von P. pastoris konnten durch die Verwendung eines einfachen und skalierbaren Isolationsprotokolls, bestehend aus Dialyse und Kationenaustauschchromatographie, zwei aktive Konformationen des Enzyms isoliert werden. Die Produktivität der THCAS wurde stark durch die Kinetik und die physikalischen und chemischen Eigenschaften der Reaktanden (Enzym und Cannabinoide) beeinflusst und konnte dennoch zwanzigfach durch eine Optimierung der Substratlöslichkeit und der Proteinkonzentration gesteigert werden. Hier erwies sich vor allem die Etablierung eines Zweiphasensystems mit Hexan als zweiter organischer Phase als vorteilhaft. Eine quantitative Prozessevaluierung zeigte, dass die erreichte Produktivität und Produktkonzentration von THCA mit industriellen Prozessen zur Herstellung von Feinchemikalien und Pharmazeutika vergleichbar ist. Die Arbeit legt eine Basis für die zellfreie biokatalytische Produktion von komplexen und hydrophoben Spezialitäten wie z.B. THCA im Gramm Maßstab.
